ABSTRACT Lettuce seeds usually show germination problems that can be related to dormancy and or thermoinhibition, as well as to the genotype constitution. The knowledge of the temperature at which the thermoinhibition process begins, as well as the influence of the age of the seed on its germination, is extremely important to establish more suitable parameters for the selection of higher genotypes in breeding programs. The objective of this work was to evaluate the seed germination of three lettuce cultivars according to the enviromental temperature and the age of the seeds. The cultivars Everglades (tolerant to thermoinhibition), Luisa (medium tolerant) and Verônica (sensitive) were used. Seeds of each cultivar were evaluated by the standard germination test, first count test, germination speed index and germination test of remaining seeds. The analyses were performed at four environmental temperatures and in seven storage periods after harvest. None of the cultivars showed primary dormancy. The tolerance to thermoinhibition showed by cultivars Everglades and Luisa and the sensitivity of cultivar Verônica were confirmed. The most suitable temperature for differentiating tolerant and sensitive thermoinhibition genotypes occurred from 30.5 ºC and 260 days after harvest.

Abstract: At higher temperatures, lettuce seeds may not germinate, resulting in problems for crop establishment in the field and production. This study aimed to evaluate the maternal effect for thermotolerance on lettuce seeds and find enzymatic markers. We used the thermoinhibition tolerant cultivar ‘Everglades’, the sensitive ‘Verônica’, their reciprocal hybrids, F1 and F2 generation. Seeds were submitted to germination and enzymatic tests (catalase, superoxide dismutase, ascorbate peroxidase, and endo- β-mannanase). Germination (first and final counting) and speed index were compared through Tukey’s test. ‘Everglades’ and its reciprocal hybrids, have not been influenced by temperature regarding germination compared to the other treatments. Also, the maternal effect was observed for the F1 generation. Higher temperatures interfere with the activity of all enzymes evaluated, consequently in the physiological parameters. However, superoxide dismutase and endo- β-mannanase have shown direct and more expressive correlation with changes on the physiological parameters compared to the other enzymes. Thus, it is possible to conclude that those enzymes can be used as markers for thermotolerant genotype selection.

Resumo: Sob altas temperaturas, sementes de alface não germinam, resultando em problemas para o estabelecimento da cultura em condição de campo. Esse trabalho analisou a possibilidade de presença de efeito maternal na termotolerância em sementes de alface e buscou marcadores enzimáticos para tal. Usaram-se as cultivares Everglades (tolerante) e ‘Verônica’ (sensível), bem como seus híbridos recíprocos e a geração F2. As sementes foram analisadas quanto à germinação, bem como atividade enzimática (catalase, superóxido dismutase, ascorbato peroxidase e endo-β-mananase). O percentual de germinação; (primeira contagem e final), bem como a velocidade (IVG) foram analisados por meio de ANOVA e teste de Tukey. A cultivar Everglades e seus híbridos recíprocos não foram influenciados pela germinação em alta temperatura quando comparados com os outros tratamentos. Além disso, o efeito maternal foi observado na geração F1. Altas temperaturas interferem na atividade de todas as enzimas analisadas nesta pesquisa, consequentemente também nos parâmetros fisiológicos. Contudo, a superóxido dismutase e a endo-β-mananase tiveram suas variações altamente correlacionadas com as mudanças nos parâmetros fisiológicos. O presente trabalho evidenciou o efeito materno para a termotolerância em sementes de alface, bem como o uso da superóxido dismutase e endo-β-mananase como marcadores enzimáticos para tal característica.

Abstract The objective of this work was to select sweet potato (Ipomoea batatas) genotypes with leaves with potential to be used for human consumption. Twenty-six experimental genotypes and four commercial cultivars were tested in a randomized complete block design with three replicates and ten plants per plot. The trial was carried out in the municipality of Lavras, in the state of Minas Gerais, Brazil. Leaf shape and lobe, aerial-part yield, edible-leaf yield, edible-leaf percentage, edible-leaf dry mass, and latex production, as well as leaf total chlorophyll, pH, soluble solids contents, titratable acidity, total phenolic compounds, and anthocyanin contents, were evaluated. Roots were characterized as to their pulp and peel colors. Fifteen genotypes presented the highest aerial-part (29.6 to 51.8 Mg ha-1) and edible leaf (7.8 to 12.7 Mg ha-1) yields. In the biochemical analysis, high contents of chlorophyll, total phenolics, and anthocyanins were observed. In addition, pH, soluble solids, and leaf titratable acidity did not differ significantly among the evaluated genotypes. Leaves of ten genotypes are recommended for human consumption.

Resumo O objetivo deste trabalho foi selecionar genótipos de batata-doce (Ipomoea batatas) com folhas com potencial para serem usadas para consumo humano. Vinte e seis genótipos experimentais e quatro cultivares comerciais foram testados em delineamento de blocos ao acaso, com três repetições e dez plantas por parcela. O ensaio foi realizado no município de Lavras, no estado de Minas Gerais, Brasil. Foram avaliados formato e lóbulo da folha, produção da parte aérea, produção de folhas comestíveis, percentagem de folha comestível, massa seca de folha comestível e produção de látex, bem como clorofila total, pH, conteúdo de sólidos solúveis, acidez titulável, compostos fenólicos totais e teor de antocianinas das folhas. As raízes foram caracterizadas quanto às cores da sua polpa e casca. Quinze genótipos apresentaram as maiores produtividades de parte aérea (29,6 a 51,8 Mg ha-1) e de folhas comestíveis (7,8 a 12,7 Mg ha-1). Nas análises bioquímicas, foram observados altos teores de clorofila, compostos fenólicos totais e antocianinas. Além disso, pH, sólidos solúveis e acidez titulável foliar não diferiram significativamente entre os genótipos avaliados. As folhas de dez genótipos são recomendadas para alimentação humana.

Abstract The objective of this work was to evaluate the occurrence of the maternal effect on thermotolerance in lettuce (Lactuca sativa) seeds, as well as to find an enzymatic marker for this trait in lettuce seedlings. Seeds from the Everglades thermotolerant cultivar, the Verônica susceptible cultivar, and their reciprocal hybrids were used. For each cultivar and reciprocal hybrid, germination percentage and vigor (germination speed index) tests were carried out at 20 and 32°C. Thermotolerance was defined as the capacity to germinate at 32°C. To obtain markers that could be associated with thermotolerance, enzymatic tests were performed. The bands of the electrophoretic enzymatic extracts were quantified using the ImageJ software. The hybrids behaved similarly to their female genitors. When cultivar Verônica was the female genitor, hybrid seeds did not germinate at high temperatures and showed a lower expression of the esterase enzyme. However, when the female genitor was the Everglades cultivar, hybrids showed a high germination at high temperatures and a greater esterase expression. Therefore, there is a maternal effect on tolerance to thermoinhibition in lettuce seeds, and the esterase enzyme can be used as a selective marker to identify homozygous progenies for this trait.

Resumo O objetivo deste trabalho foi avaliar a ocorrência do efeito materno na tolerância à termoinibição em sementes de alface (Lactuca sativa), bem como encontrar um marcador enzimático para esta característica em mudas de alface. Foram utilizadas sementes da cultivar termotolerante Everglades, da cultivar suscetível Verônica e de seus híbridos recíprocos. Para cada cultivar e híbrido recíproco, foram conduzidos testes de germinação e vigor (índice de velocidade de germinação) a 20 e 32°C. A termotolerância foi definida pela capacidade de germinação a 32°C. Para encontrar marcadores que pudessem ser associados com a termotolerância, foram realizados testes enzimáticos. As bandas eletroforéticas dos extratos enzimáticos foram quantificadas por meio do programa ImageJ. Os híbridos apresentaram comportamento semelhante ao dos seus genitores femininos. Quando a cultivar Verônica foi o genitor feminino, as sementes dos híbridos não germinaram sob alta temperatura e apresentaram menor expressão da enzima esterase. Porém, quando a cultivar Everglades foi o genitor feminino, os híbridos apresentaram alta germinação sob temperatura elevada e maior expressão da esterase. Portanto, há efeito materno na tolerância à termoinibição em sementes de alface, e a enzima esterase apresenta potencial como marcador para identificar progênies homozigotas para essa característica.

Resumo: A resposta germinativa de sementes submetidas à condição de estresses artificiais são ferramentas para um melhor entendimento da capacidade de sobrevivência e adaptação das espécies em condições de estresses naturais. Diante disso, o objetivo neste trabalho foi avaliar o efeito do condicionamento fisiológico na germinação de sementes, quebra de dormência e comprimento de plântulas de Urochloa brizantha sob condições de restrição hídrica e estresse salino. O delineamento utilizado foi inteiramente casualizado, com quatro repetições, em esquema fatorial (3 x 6), sendo três condições de germinação (água, restrição hídrica com PEG e salinidade com NaCl) e 6 tratamentos que constituíram as cinco soluções condicionantes (água destilada, nitrato de potássio, espermidina, giberelina e nitroprussiato de sódio) mais um controle (semente não condicionada). Determinou-se: teor de água após condicionamento (antes da secagem) e após a secagem das sementes; germinação aos sete e aos vinte e um dias após a semeadura; índice de velocidade de germinação; viabilidade pelo teste de tetrazólio das sementes remanescentes do teste de germinação; e comprimento de plântulas. O condicionamento fisiológico é eficiente em minimizar os efeitos negativos da restrição hídrica e salinidade. Com a utilização de nitroprussiato de sódio no condicionamento ocorre menor porcentagem de sementes dormentes e maior germinação, vigor e desenvolvimento de plantas.

Abstract: Analyses of germination of seeds under artificial stress conditions are tools for a better understanding of the survival and adaptation capacity of species under natural stress conditions. Therefore, the aim of this study was to evaluate the effect of priming on seed germination, on breaking of dormancy, and on the length of Urochloa brizantha seedlings under water restriction and salt stress conditions. A completely randomized experimental design was used with four replications in a 3 × 6 factorial arrangement consisting of 3 germination conditions (water, water restriction with PEG, and salinity with NaCl) and 6 treatments [five priming solutions (distilled water, potassium nitrate, spermidine, gibberellin, and sodium nitroprusside) and a control (unprimed seeds)]. The following determinations were made: seed moisture content after priming (before drying) and after drying the seeds, germination test at seven and twenty-one days after sowing, germination speed index, viability by the tetrazolium test of the seeds remaining from the germination test, and seedling length. Priming is effective in minimizing the negative effects of water restriction and salinity. Use of sodium nitroprusside as a priming solution results in a lower percentage of dormant seeds and greater germination, vigor, and plant development.

Resumo O objetivo deste trabalho foi determinar o controle genético da tolerância à termoinibição em sementes de alface (Lactuca sativa). Utilizaram-se sementes das gerações F1, F2 e F2:3 oriundas do cruzamento entre as cultivares Everglades (tolerante à termoinibição) e Verônica (sensível à termoinibição), além de sementes dos próprios genitores. Realizaram-se testes de germinação das sementes em delineamento completamente casualizado, com quatro repetições para cada um dos genitores, oito para a F1 e quatro para cada uma das 26 progênies F2:3. A média da população F2 foi considerada como a média da amostra das 26 progênies. O controle genético da tolerância à termoinibição em sementes de alface é atribuído a um ou poucos genes. Os efeitos aditivos são mais expressivos do que os não aditivos, e a herdabilidade no sentido restrito é relativamente alta, o que permite antever sucesso com a seleção.

Abstract The objective of this work was to assess the genetic control of tolerance to thermoinhibition in lettuce (Lactuca sativa) seeds. Seeds of the F1, F2, and F2:3 generations derived from the cross between cultivars Everglades (tolerant to thermoinhibition) and Verônica (sensitive to thermoinhibition) were used, besides seeds from the parents. Seed germination tests were conducted in a completely randomized design, with four replicates of each parent, eight of F1, and four of each of the 26 F2:3 progenies. The mean of the F2 population was considered as the mean of the sample of the 26 progenies. The genetic control of thermoinhibition tolerance in lettuce seeds is attributed to one or a few genes. The additive effects are more expressive than the nonadditive ones, and narrow-sense heritability is relatively high, allowing the prediction of success in selection.

Abstract In comparison to tomato plants without the dwarfism gene, dwarf tomato plants were shorter, had a smaller canopy diameter and remained upright during maturation, thereby reducing the fruit-soil contact, resulting in healthier fruits. This study evaluated tomato families of a BC1F3 population carrying the dwarf gene for industrial yield. The 150 BC1F3 families and the two parents (hybrid H-9889 and accession BGH-9889) as controls were assessed in an augmented block design with four replications. The data were subjected to variance analysis and the F test. A selection index, based on the sum of ranks of Mulamba and Mock (1978), was used for the selection of genotypes, considering the most relevant traits, i.e., canopy diameter, yield, lodging and fruit firmness. Thus, selection resulted in a yield gain of 8.04%.
